초록

Extracellular Vesicles (EVs) are nano-sized particles composed of lipid bilayer. It contains proteins, lipids, and nucleic acids regulating cellular protein expression and function. This study aimed to extend EVs research focused on terrestrial organisms by exploring marine- derived sources for potential therapeutic applications. Accordingly, we investigated the effects of octopus derived extracellular vesicles(OEVs) on wound healing through the evaluation of their anti-inflammatory activity and influence on mesenchymal stem cells (MSCs) migration. Octopus tissues were decellularized and enzymatically digested, followed by tangential flow filtration(TFF) to isolate EVs smaller than 200 nm. The extracted OEVs were characterized and assessed for effects on cell proliferation, cytotoxicity, inflammation and MSCs migration. Physical properties consistent with previously reported EVs were observed. PKH-26 labeled OEVs were efficiently taken up by cells through endocytosis. Moreover, OEVs treatment enhanced the proliferation of RAW 264.7 macrophages and MSCs without cytotoxicity. In addition, OEVs suppressed LPS-induced inflammatory responses and significantly promoted MSCs migration compared to control. These findings reveal that OEVs exert significant anti-inflammatory and pro-regenerative effects. Therefore, we suggest that OEVs have potential as a novel marine-derived therapeutic candidate for wound healing
