Alternative testing methods have gained attention due to ethical concerns and the variability of animal testing. The cornea, as a transparent barrier tissue of the eye, is highly susceptible to inflammation such as keratitis. Therefore, establishing an in vitro cornea model that can accurately reproduce inflammatory responses is crucial for evaluating drugs. For this reason, we fabricated a rabbit cornea model by culturing self-aggregated rabbit corneal keratocytes (rbCKs) in trans-well inserts and subsequently seeding rabbit corneal epithelial cells (rbCECs) on the aggregates. The model was characterized with matrix formation, live/dead staining, and histological observation. To induce the inflammatory condition, cytokines were applied, and the response to Ciprofloxacin (CIP) was assessed by cell viability and Western blot analysis. A low concentration of CIP alleviated inflammatory responses, whereas higher concentrations increased the expression of pro-inflammatory cytokines. These results demonstrate that the inflammationinduced model can be utilized to evaluate the efficacy of drugs. From a future perspective, 3D bioprinting is expected to reproduce native collagen architecture better and enhance test reproducibility in the model.
