Abstract
Background: As the number of LC-MS/MS tests with high accuracy and precision have increased, sample pretreatment methods have also be- come more complex, thus making them more difficult for analysts to perform them manually. To overcome this, we implemented Janus G3 Auto- mated Workstations (Revvity, Turku, Finland) and developed an automated sample pretreatment method for LC-MS/MS based quantitation of plas- ma metanephrine and normetanephrine in this study. Methods: The feasibility and analytical performances of the method including precision, accuracy, linearity, and carryover, were analyzed and compared with that of the manual sample preparation method. Results: The coefficients of variations for repeatability and within-laboratory precision were less than 10% for metanephrine and normetaneph- rine assays, meeting acceptable criteria. Accuracy bias values were less than 5%, also within acceptable limits. The verified linear ranges were 0.02 to 95.07 nmol/L for metanephrine and 0.02 to 102.34 nmol/L for normetanephrine. The pretreatment step alone using the Janus G3 Automat- ed Workstations took 45 minutes, significantly shorter than the manual pretreatment method. The comparison between manual and automated pretreatment methods of LC-MS/MS measurement showed excellent agreement, with the following results: a correlation coefficient of 0.9958, slope of 1.000 (1.000–1.031), and intercept of 0.000 (-0.006–0.000) for plasma metanephrine, and 0.9976, 1.000 (0.972–1.005), and 0.000 (-0.002– 0.022) for plasma normetanephrine, respectively. Conclusions: This automated sample pretreatment method with Janus G3 Automated Workstations showed acceptable performances for pre- treatment steps of LC-MS/MS quantitation regarding both metanephrine and normetanephrine. This method offers a beneficial solution for ana- lysts by reducing labor.
