초록
Objectives: This study was designed to examine apoptosis and metastasis inhibition effects of Melonis Pedicellus (MP) extracts on breast cancer. Methods: Apoptosis and cell migration inhibition effects were tested with 4T1 cells. To investigate apoptosis, we measured cell cytotoxicity, reactive oxygen species (ROS), and morphological change of cell, mitochondria, and cell nucleus. Also, we measured expression of Ki 67 to investigate the inhibition of cell proliferation. To investigate cell migration inhibition effects, we conducted wound healing assay and analysis of matrix metalloproteinase-2 (MMP2) and MMP9 mRNA expression. At last, we analyzed phosphorylation of extracellular signal regulated kinase (P-ERK) and phosphorylation of p38(P-p38) expression to verify the mechanism of apoptosis. Results: After treating 4T1 cells with 25, 50, 75, 100, 250, 500 μg/ml of MP, cell toxicity and morphological change of cell were observed compared to the control group. Also, when 4T1 cells were treated with 500 μg/ml of MP, ROS expression, mitochondria fission, condensation of cell nuclei, and decreased expression of Ki 67 were observed. After treating 4T1 cells with 500 μg/ml of MP and conducted scratch wound healing, migration wound healing area was decreased compared to the control group, and mRNA expression of MMP2 and MMP9 also decreased. Furthermore, P-ERK and P-p38 expression both increased compared to the control group, but only P-p38 expression was statistically significantly increased. Conclusions: MP might have anti-cancer effects on breast cancer by apoptosis and metastasis inhibition.
