Plant abiotic stress tolerance has been modulated byengineering the trehalose synthesis pathway. However, manystress-tolerant plants that have been genetically engineered forthe trehalose synthesis pathway also show abnormaldevelopment. The metabolic intermediate trehalose 6-phosphatehas the potential to cause aberrations in growth. To avoidgrowth inhibition by trehalose 6-phosphate, we used a genethat encodes a bifunctional in-frame fusion (BvMTSH) ofmaltooligosyltrehalose synthase (BvMTS) and maltooligosyltrehalosetrehalohydrolase (BvMTH) from the nonpathogenicbacterium Brevibacterium helvolum. BvMTS converts maltooligosaccharidesinto maltooligosyltrehalose and BvMTHreleases trehalose. Transgenic rice plants that over-expressBvMTSH under the control of the constitutive rice cytochromec promoter (101MTSH) or the ABA-inducible Ai promoter(105MTSH) show enhanced drought tolerance without growthinhibition. Moreover, 101MTSH and 105MTSH showed anABA-hyposensitive phenotype in the roots. Our results suggestthat over-expression of BvMTSH enhances drought-stresstolerance without any abnormal growth and showes ABAhyposensitive phenotype in the roots.

Moreover, 101MTSH and 105MTSH showed anABA-hyposensitive phenotype in the roots. Our results suggestthat over-expression of BvMTSH enhances drought-stresstolerance without any abnormal growth and showes ABAhyposensitive phenotype in the roots.
