초록

Understanding molecular characteristics and metabolicprocesses of the mammalian endometrium is crucial for advancingbiological research, particularly in veterinary obstetricsand pathology. This study established and analyzed organoidsfrom endometrial epithelial stem cells of five mammals withdifferent placental types: cows (cotyledonary), dogs and cats(zonary), pigs (diffuse), and rats (discoid). Organoids from thesefive species were maintained for over 13 passages, frozen, andthawed. Pathological analysis confirmed that they retained characteristicsof their original tissues. Furthermore, integrative transcriptomeanalysis of organoids and tissues from the five specieshighlighted key pathways such as PI3K–Akt signaling and extracellularmatrix–receptor interaction known to be crucial in cancerresearch. Although genes associated with vascular smooth musclecontraction were downregulated, these organoids exhibited significantactivities of genes involved in hormone metabolism. Inconclusion, our study achieved stable establishment of endometrialorganoids from five mammals with different placental types, offering foundational data for organoid research. In the future, theseorganoids are suitable models for investigating uterine physiologyand diseases and for developing potential therapies.
