Abstract
We want to provide one of basis for the control of NHOK (Norm머 Human Oral Keratinocyte) proliferation and regeneration by the assessment the effect of NMDA (N-methyl-D-as맹πate) and NOS-1 (Ni머c Oxide Synthase Inhibitor) on NHOK proliferation with the quantitatively evaluation of not only the living cell number but 외so the DNA synthesis. The NHOK were c띠tured from the gingival tissue which were obtained by 20 person (the age from 20 to 29), and the P3 cell were used for this study. The viability were assessed by the MTT assay and the DNA synthesis were evaluated by the BrdU assay. NMDA and NNA (L-nitroarginine; inhibitor of ne따"()nal and endothelial NOS, calcium dependent) inducE영 living cell number in early stages and they produced the decreasing of the living cell number in latter period. However, the NAME (L-nitr,∞rginine methyl ester; the methyl ester form of NNA and non selective NOS inhibitor, calcium independentl did not produce the decreasing of the living cell number in latter period. Therefore, we assumed that the NMDA effect of the decreasing the NHOK cell number in long term period is owing to the calcium ion change. In the meanwhile, the effects of the media changing and the drugs on the DNA synthesis had been occurred within sever잉 hours, and that effects were disappeared before 24 hours.
