Abstract
Until nuw, several factors have bæn examined alune or in combination, to be effective on cell proliferation, differentiation and chemotaxis which are connected with periodontal tissue healing and regeneration. One of them, dimethyl sulfoxide (DMSO) is widely used as a solvent for molecules that are relatively insoluble in water or other solvents and is also used for cryoprotection in the fræzing of living cells. DMSO shows a wide range of primary ph81macological actions according to the type of celis, concentrations, incubation media and so on. However, there are few or no reports abcut effects of DMSO for primary cultured cells of periodontal cells. Therefore, in this study we examined apoptosis of NHOK and the inhibition effects of DMSO on the proliferation of normal human periodontal cells with vru;ous concentrations. Apoptosis in the NHOK was examined by Westem blot analysis and TUNEL labeling test. NHOK were cultured with in the absence {contro]) or presence of different concentrations of DMSO for 1 day. Cell proliferation was assessed by MTT assay Normal human gingiv머 fibroblasts, normal human periodontalligament cells and MG 63 cells were cultured in the absence (contro]) or presence of different concentrations of DMSO for 1, 2, 3 and 7 days 1n NHOK, typical features of apoptosis were maJlÏfested at 4% group. Cleaved caspase ~3 was shown in Westem blot analysis and labeling of nucleus was shown in TUNEL labeling test. 1n MTI' assay test, DMSO caused statistically significant decrease (P < 0.001) compared to controls in normal human gingival fibroblasts and normal hurnan periodontal ligament cells after 2 days at 2% group, and in MG 63 cells after 1 day at 2% group. The results from this study suggest that the DMSO with 2% can decrease the proliferation of periodontal cells without apoptosis ín vitro. These results provide useful information further in vivo studv abcut the application of DMSO for dental treatrnents
