Abstract
The diagnosis of Mycobacterium tuberculosis( M tuberculosis) in body fluids is not easy due to their small copy numbers and several technical problems. Recently molecular diagnosis for M. tuberculosis by Polymerase chain reaction(PCR) has frequently been used. In this study, the nested PCR method was tried to obtain higher sensitivity for the detection of M. tuberculosis and compared with the first step PCR in a total of 400 specimens. The oligonucleotide sequences of primers were selected from IS6110 gene of M. tuberculosis and were designed to amplify a 324 bp for first PCR and a 239 bp for Nested PCR, respectively. In 400 body fluid samples, 82( 20.5 %) cases were positive in the first PCR group. However, in Nested PCR group, 190(47.5 %) cases were positive. Sensitivity of Nested PCR group was more than doubled. This study showed that the nested PCR method has higher sensitivity than first PCR. Therefore, it is our opinion that nested PCR, especially in body fluids , is more useful tool for diagnosis of M. tuberculosis than first PCR method.
