Abstract
In order to study on the expression and function of human folate receptor(hFR) in ovarian carcinoma cell line (IGROV1), Nasopharyngeal epidermoid carcinoma cell line (KB), breast carcinoma cell line(MCF-7), and brain cancer cell line(GH3) , I isolated hFR cDNA from human placenta cDNA library and constructed a recombinant plasmid inserted the hFR cDNA into pGM4Z plasmid. I carried out Western blot, Northern blot, cell surface binding assay, and transport assay to compare the expression and function of hFR in cancer cell lines each other. I discovered that IGROV1 and KB cell lines express the high level of hFR, cell surface binding activity, and transport ability, but MCF-7 and GH3 cell lines do not express detectable level of hFR. Consequently, it’s possible to use the amount of expression of hFR to diagnose a ovarian carcinoma and nasopharyngeal epidermoid carcinoma from other cancers.
