Abstract

The study was focused on the estimation of genetic diversity of Helicobacter pylori. Thirty strains were isolated from 90. gastric ulcer tissues. The strains were indentified by typical Gram stain and conventional biochemical tests. Antral biopsies were taken for culture from 90 patients with chronic gastritis, gastric ulcer and duodenal ulcer at the time of endoscopy. H. pylori was identified by typical gram stain morphological and biochemical tests. UreA of the candidates were Amplified using PCR, and 30 strains were identified as H. pylori from 90 specimens. Repetitive sequence based polymerase chain reaction (Rep-PCR) fmgerprinting was perfonned by primers BOXA1R (5’-CTACGGCAAGGCGACGCTGACG-3’). Isolates were divided into five molecular types (1 -V) at a similarity value of 0.5 using Repetitive Sequence-Based Polymerase Chain Reaction fmgerprinting. Distribution of the similarity value was 0.10 to 1.00, which shows a wide range of similarity value. DNA fmgerprinting of the 30 isolates were analyzed by Repetitive sequence based polymerase chain reaction (Rep-PCR). Isolates were divided into five molecular types (1 -V) at a similarity value of 0.5 using Rep-PCR fmgerprinting. Distribution of the similarity value was 0.10 to 1.00, which show a. wide range of similarity value. These results suggested that strains isolated in patients have much diversity. In conclusion, Rep-PCR fmgerprinting of H. pylori could be available for epidemoiological infections and for clinical applications.
