Abstract

This study conducted STR (Short Tandem Repeat) analysis using umbilical cord tissue that had been fixed in 4% neutral buffered formalin for approximately three months and compared the analytical efficiency with that of formalin-fixed paraffin-embedded (FFPE) samples prepared from the same tissue. The formalin-fixed tissues were pretreated by washing for 24 hours, 10 days, or 20 days, while the FFPE samples underwent xylene-based deparaffinization. The results showed that the FFPE samples yielded stable STR profiles across all loci and exhibited high DNA concentrations, whereas the formalin-fixed tissues showed limited STR profiles only after 20 days of washing, and DNA was not detected under shorter washing conditions. These findings demonstrate that paraffin embedding effectively suppresses formalin-induced DNA damage and helps maintain the efficiency of STR analysis. This study highlights the applicability of FFPE samples in STR analysis of pathological tissues and underscores the need for standardized pretreatment protocols, providing foundational data for practical use in forensic casework.
