초록
Chili peppers are one of the essential crops world-widely. Mutant populations (M2) using gammarayand carbon ion beam irradiation in pepper seeds (Yuwolcho) were established in 2016. In these populations,2,880 mutant lines were screened using the Target Induced Local Lesions IN Genomes (TILLING) method. TILLING is a reverse genetics technique that allows the identification of mutants with target-gene mutations usingrelatively straightforward experiments including PCR, heteroduplex formation, digestion by an endonuclease, andelectrophoresis. The target gene chosen for this study was the FLOWERING LOCUS T (FT) and its homologs (FTlikegenes), which is associated with flowering regulation. Chili peppers have a total of six FT and FT-like genes, and mutations in four genes were screened through TILLING. Statistical analysis of the experimental results revealedthat the probability of selecting a mutation per a gene was approximately 1.35 × 10−4. This suggests that a mutantpopulation of 7,419 individuals is required to isolate average one mutant of the desired gene. This result providesuseful information to determine the population size in mutation breeding using irradiation.
