초록
Wrinkles formed from the aging of human skin are mainly caused by intrinsic and extrinsic factors. Intrinsic aging factors include increased age and stress while extrinsic aging factors include ultraviolet rays (UV), stress, and smoking. The above factors increase ROS, an active oxygen radical, in vivo. These factors then increase MMPs, one of the intracellular proteins, to inhibit collagen synthesis, which is important for skin elasticity. As the main cause of skin aging, inhibited collagen synthesis has a significant impact on wrinkle formation. Previous studies have shown the effects of Akebia quinata on antioxidant, anti-inflammatory, anti-aging, and anti-melanin activity properties. However, there have been no details on the pathways in the human body through which MMPs can be inhibited. In this study, an MMP-1 ELISA kit was used to determine whether Akebia quinata fruit water extract (AQFWE), a fruit native to Korea, is effective in wrinkle prevention. We found that AQFWE significantly inhibits MMP-1. As a result of confirming the toxicity of AQFWE in Hs68 cells by MTT assay, it was confirmed that there was no toxicity found at any concentration. ROS scavenging ability was confirmed through DPPH assay. Real-time RT-qPCR was performed on Hs68 cells with TNF-α treatment at concentrations of 200, 400, and 800μg/mL to confirm that MMP- 1 was significantly inhibited. Furthermore, western blotting was performed at the same concentration to confirm the MMP-1 inhibitory ability of AQFWE through MAPKs, AP-1, and NF-κB pathways. This result suggests that AQFWE effectively inhibits MMP-1 and has antioxidant and anti-aging properties.
