Abstract
In the present study, DNA marker that differentiates PCNA from other non-PCNA was developed by bulked segregant analysis (BSA) using RAPDs. DNA samples obtained from 30 PCNA and 32 non-PCNA persimmon cultivars were bulked depending on the types of astringency loss. Four-hundreds RAPD primers were screened on bulked DNA samples and a non-PCNA-specific PCR band was obtained from OPJ18 primer. For marker conversion of the RAPD to simple and robust PCR-based marker, non-PCNA linked PCR band was cloned and a sequence characterized amplified region (SCAR) marker J18SCAR-280 was designed based on sequence variations. In addition, genetic linkage of J18SCAR- 280 to PCNA trait was validated by evaluating‘Jamisi’(PVNA) x‘Taishu’ (PCNA) -derived progenies segregating for the astringency trait. Although further investigations based on larger segregating populations, our results implied that J18SCAR-280 can be useful for marker-assisted selection of the PCNA trait.
