Abstract
This study is the first report of central blackening in onion bulbs caused by Pseudomonas oryzihabitans, an opportunistic pathogenin humans and warm-blooded animals. The purpose of this study is to isolate and identify the pathogen causing onion bulb blackeningand to investigate its microbiological characteristics. In 2021, a center blackening of onion bulbs that could cause consumer disgustwas observed in onion bulbs in Korea. Gram-negative, non-spore-forming, rod-shaped pseudomonads were isolated from black pigmentedtissue of onion bulbs. Symptoms after artificial infection formed through the pathogenicity assay were the same as those of naturalinfection, and there were no symptoms in the negative control treated with sterile distilled water. The causative pathogen was identified as Pseudomonas oryzihabitans through pathogenicity assay, API (analytical profile index) test, MALDI-TOF MS (whole-cell matrix-assistedlaser desorption/ionization time-of-flight mass spectrometry), and 16S rRNA gene region sequencing and phylogenetic analysis. Althoughno cases of onion damage from large-scale field or post-harvest disease have been reported, the results obtained in this study suggestthat P. oryzihabitans could potentially pose a threat to onion global production and transportation and food safety. In addition, sinceit is an opportunistic human pathogen, it can be seen that it has a risk of being infected from onions to the human body at any time.
