Abstract
Radix Astragali, (root of Astragalus) is a traditional oriental medicine used as a tonic and diuretic. Astragalus membranaceus (Fisch) Bunge and A. membranaceus (Fisch) Bunge var. mongholicus (Bunge) are the most commonly used Huangqi. For consumers, the authentic identification of the origin of a particular Astragalus species is very difficult. To overcome this problem, we developed an efficient method to discriminate from Astragalus membranaceus (Fisch) Bunge and A. membranaceus (Fisch) Bunge var. mongholicus (Bunge). The specific repeatable markers were designed by the cloning of significant RAPD bands obtained in PCR. Seventeen specific RAPD bands were obtained from 40 OPERON primer sets. Two out of 17 RAPD bands were cloned into pGEM-T-EASY vector and then subjected to the nucleotide sequence analysis. In particular, the random primer C18 (5'-TGAGTGGGTG-3'), which showed polymorphism in Astragalus species, amplified 832 bp products, which was specific to Astragalus membranaceus (Fisch) Bunge. Based on nucleotide sequence of the primer C18 product, we designed a JIN-Q3 (5'-CGCTACAACCTTGCTTTAAT-3') and JIN-4(3'-AGAACATAAACTTTCCTCATGTA- 5') primer-set to amplify a 434 bp sized SCAR (sequence characterized amplified regions) marker, through which Astragalus membranaceus (Fisch) Bunge and A. membranaceus (Fisch) Bunge var. mongholicus (Bunge) can be efficiently discriminated.
