Abstract
plifiedtechniquewhichsimultaneouslyinducesandcryoprotectsembryogeniccalli usingplantvitrificationsolution2(PVS2)fol-lowedbydehydrationwasdevelopedforthecryopreservationofSoapberygeneticresources. VitrificationisawayofremovingthemoistureinvegetationthroughPVS2.ThePVS2vitrificationsolutionconsistedof30%glycerol(w/v),15%ethyleneglycol(w/v),15%Dimethylsulfoxide (w/v) in B5 medium containing 0.4Msucrose. Two tests were done. The one was to eliminate moisture at℃ ℃ survivalratewashigherat 0 thanat25 .Inparticular, thesurvivalratereachedmorethan80%. Water-damagedembryosturned℃ ℃brown and stoped growth, but energetic embryos took on a milky hue and show a very vigorous growth rate. Successful cry-opreservationofsomaticembryosof soapberrycanbeusedtoestablishinvitrogenebanksforlong-termconservationofSoapberrygenetic resources to complement field genebanks and otherin vitromethods already being used.Key words- Cryopreservation, Somatic embryo,Sapindus mukorosi, Vitrification
