초록

Vibrio tapetis, originally known as the causative agent of brown ring disease in clams, has recently emerged as an opportunistic pathogen of olive flounder (Paralichthys olivaceus), with recurrent de- tections during low-temperature seasons. To address this threat, we developed a formalin-killed cell (FKC) vaccine and two adjuvanted formulations using squalene–aluminum hydroxide (FKC-SQ) or Montanide™ ISA 760 VG (FKC-Seppic). Vaccinated olive flounder were assessed for antibody re- sponses, histopathological changes, and protective efficacy. FKC-Seppic induced the highest agglutinat- ing antibody titers, whereas ELISA showed uniformly elevated antibody levels across all vaccine groups. Histopathology revealed severe granulomatous inflammation and organ adhesions in FKC-Seppic fish, while FKC-SQ caused only mild localized inflammation and FKC minimal tissue reactions. After challenge with V. tapetis at 15℃, survival reached 83.33% (RPS 66.67%) in both adjuvanted groups and 72.22% (RPS 44.44%) in the FKC group, compared with 50% in control. Although differences were not statistically significant, vaccinated fish consistently exhibited improved survival. These findings indicate that FKC-based vaccines provide partial protection against V. tapetis, and that the squalene–alum formulation offers a more favorable balance of efficacy and safety than Montanide ISA 760 VG, supporting its potential application in olive flounder aquaculture.
