Clozapine은 비전형적인 항정신병 약물로서 세계적으로 점차 사용이 증가되고 있다. Clozapine은 임상적 효능, 부작용, 작용 기전 등에서 전형적인 항정신병 약물과는 구별된다. 이런 특성으로 인해 정신분열증의 원인을 더욱 밝히고 더욱 선택적으로 작용하는 새로운 항정신병 약물을 개발할 수 있는 가능성을 제시해 준다. 그러나 다른 항정신병 약물에 비해 치명적인 부작용인 무과립구증의 높은 발생율 때문에 사용이 제한되어 왔다. 그래서 clozapine을 효과적이고 효율적으로 사용하기 위해서 임상의는 clozapine으로 발생 가능한 이득과 위험성을 알고 있어야 한다.
Abstract
Clozapine is an atypical antipsychotic drug being used with increasing frequency throughout the world. The clinical efficacy, side effects, and action mechanism of clozapine differentiate it from typical antipsychotics. Its unique properties has given us hope that we can gain insight into etiology of schizophrenia and new antipsychotics which has more selective properties will be developed. Its development and use have been limited by its higher frequency, compared with other pharmacologic treatments, of the potentially fatal side effect of agranulocytosis. Thus in order to use clozapine most effectively and efficiently, clinicians must be aware of its potential benefits and risks.
