Abstract

Object : This study was designed to evaluate the effects of alcohol anticraving agent (acamprosate) to the expression of tyrosine hydroxylase(TH) in the ventral tegmental area and substantia nigra in rats with chronic ingestion of 5% ethanol by immunohistochemistrical measures. Methods : Adult Spraque-Dawley rats were used. They were assigned to ethanol exposure group(N=10) and normal control without ethanol exposure(N=10). After 4 weeks of ethanol exposure, the ethanol exposure group rats were injected 400mg/kg/i.p. of acamprosate for 3 weeks. After completing the 3 weeks of acamprosate and vehicle injection, rats were anesthetized with pentobarbital sodium (50mg/kg/ip) and perfused with 4% paraformaldehyde in 0.1 M phosphate buffer(PB), pH 7.4. The brains were removed and postfixed in the same fixative overnight, then frozen sections of 40uM thickness were made in the coronal plane. Sections were stained for detection of TH according to the immunohistochemical method. RESULTS : 1) The ethanol exposure group revealed the increased tyrosine hydroxylase expression in the VTA and substantia nigra as compared with normal control group. 2) After daily injection of acamprosate for 3 weeks, the ethanol exposure group showed decrease of tyrosine hydroxylase expression in those areas. CONCLUSION : Above findings suggest that chronic exposure to ethanol activate the dopaminergic systems in the VTA and substantia nigra. Reversely, the chronic administration of acamprosate suppresses the TH expression. It mean that the action mechanism of acamprosate via GABA and glutamate receptor eventually affect to the dopaminegic system.
