Comparative Proteome Analysis of Seed Storage Proteins in Allogamous and Autogamous Buckwheat

Abstract

Seed proteins of allogamous buckwheat (Fagopyrum esculentum Moench. cv. Miyazakizairai) and autogamous buck-wheat were separated by two-dimensional gel electrophoresis (2-DE) and characterized by gasphase sequencing. Total of 100 pro-could be used as markers for the identification of autogamous cultivar. Randomly selected 17 proteins were subjected to Nterminalsequencing. Nterminally blocked proteins (~80%) were subjected to Cleveland peptide maping followed by microsequencing.The sequences obtained were compared with those of the SWIS-PROT and NCBI databases. A protein showed high sequencehomology with 22 kDa allergen protein of buckwheat and seven with proteins such as allergen Ara H1, cysteine proteinase 2, ribo-Arabidopsis, peanut and maize, respectively. In autogamous buckwheat, the N-ter-minal sequence of three proteins showed homologous to glutelin type-A II precursor, glutelin type-B 2 precursor and putative 30Sribosomal protein S31 from rice.
