We investigated the effects of β-adrenergic activation on bonemarrow adiposity and on adipogenic differentiation of bonemarrow mesenchymal stem cells (BMSCs). C57BL/6 mice weresubjected to a control (CON), high calorie (HIGH) or lowcalorie (LOW) diet for 12 weeks. In each group, mice weretreated with vehicle (VEH) or propranolol. The number ofadipocytes per area bone marrow was increased in LOWVEHand HIGHVEH mice compared with CONVEH mice, whichwas attenuated by propranolol. Isoproterenol increased lipiddroplet accumulation and adipogenic marker gene expressionin 3T3-L1 preadipocytes and mouse BMSCs, which wereblocked by propranolol. Conditioned medium obtained fromMC3T3-E1 osteoblasts suppressed adipogenic differentiation of3T3-L1 cells, which was significantly attenuated by treatmentof MC3T3-E1 cells with isoproterenol. These data suggest thatβ-adrenergic activation enhances bone marrow adipogenesisvia direct stimulation of BMSCs adipogenesis and indirectinhibition of osteoblast anti-adipogenic potential.
