Cell proliferation is a delicately regulated process that couplesgrowth signals and metabolic demands to produce daughtercells. Interestingly, the proliferation of tumor cells immenselydepends on glycolysis, the Warburg effect, to ensure a sufficientamount of metabolic flux and bioenergetics for macromoleculesynthesis and cell division. This unique metabolic derangementwould provide an opportunity for developing cancer therapeuticstrategy, particularly when other diverse anti-cancer treatmentshave been proved ineffective in achieving durable response, largely due to the emergence of resistance. Recent advances indeeper understanding of cancer metabolism usher in newhorizons of the next generation strategy for cancer therapy. Here, we discuss the focused review of cancer energymetabolism, and the therapeutic exploitation of glycolysis andOXPHOS as a novel anti-cancer strategy, with particularemphasis on the promise of this approach, among other cancermetabolism targeted therapies that reveal unexpectedcomplexity and context-dependent metabolic adaptability, complicating the development of effective strategies.
