Mitochondria are important organelles that regulate adenosinetriphosphate production, intracellular calcium buffering, cellsurvival, and apoptosis. They play therapeutic roles in injuredcells via transcellular transfer through extracellular vesicles, gap junctions, and tunneling nanotubes. Astrocytes can secretenumerous factors known to promote neuronal survival, synaptic formation, and plasticity. Recent studies have demonstratedthat astrocytes can transfer mitochondria to damagedneurons to enhance their viability and recovery. In this study,we observed that treatment with mitochondria isolated fromrat primary astrocytes enhanced cell viability and amelioratedhydrogen peroxide-damaged neurons. Interestingly, isolated astrocyticmitochondria increased the number of cells under damagedneuronal conditions, but not under normal conditions, although the mitochondrial transfer efficiency did not differbetween the two conditions. This effect was also observed aftertransplanting astrocytic mitochondria in a rat middle cerebralartery occlusion model. These findings suggest that mitochondriatransfer therapy can be used to treat acute ischemic strokeand other diseases.
