초록
DNA methylation is one of the most extensively studied epigeneticregulatory mechanisms, known to play crucial roles invarious organisms. It has been implicated in the regulation ofgene expression and chromatin changes, ranging from globalalterations during cell state transitions to locus-specific modifications. 5-hydroxymethylcytosine (5hmC) is produced by amajor oxidation, from 5-methylcytosine (5mC), catalyzed by theten-eleven translocation (TET) enzymes, and is gradually beingrecognized for its significant role in genome regulation. Withthe development of state-of-the-art experimental techniques, ithas become possible to detect and distinguish 5mC and 5hmCat base resolution. Various techniques have evolved, encompassingchemical and enzymatic approaches, as well as thirdgenerationsequencing techniques. These advancements havepaved the way for a thorough exploration of the role of 5hmCacross a diverse array of cell types, from embryonic stem cells(ESCs) to various differentiated cells. This review aims to comprehensivelyreport on recent techniques and discuss the emergingroles of 5hmC.
