Limited efficacy and severe side effects often result insuboptimal outcomes to solid tumor therapies. In contrast, thereduced side effects and potential long-term benefits of tumorimmunotherapy offer promise, notwithstanding the challengesof variable patient responses and immune-related adverseevents hindering its widespread application. Recent advancesin mRNA technology have revolutionized cancer immunotherapy. The versatility of mRNA as a vaccine and therapeutic agent isevident in it overcoming the limitations of traditional approachesby reducing in vivo toxicity and enhancing immuneresponse activation. The synergy between mRNA technologyand immunotherapy is increasingly being utilized to improvecancer treatment efficacy. One critical aspect of maximizingthe therapeutic impact of mRNA-based treatments is the selectionof an effective delivery system. Due to their size propertiesand material characteristics, nanoparticles offer a transformativesolution, enabling the targeted and efficient delivery ofmRNA to tumor tissues or immune cells. This precision deliverymechanism significantly enhances the effectiveness of immunotherapy, and represents a significant advance in cancertreatment. This review aims to explore how mRNA delivery viananoparticles enhances tumor immunotherapy. Examination ofits applications and challenges provides insights and strategicperspectives to advance this innovative therapeutic approach.
