Abstract

We have previously reported that intraperitoneal (i.p.) injection of Korean red ginseng water extract demonstrated preventive and therapeutic effects on testicular toxicity induced by 2,3 ,7,8-tetrachorodibenzo-p-dioxin (TCDD) in young male guinea pigs. Further study was carried out to fmd the active compound(s) and to confmn the protective effect in female guinea pigs by determining hematological parameters. A total of 80 female adult guinea pigs (800 ± 20g) were divided into 8 groups: group 1 (normal control group) received vehicle (corn oil containing small amount of acetone and DMSO) and saline; group 2 (single TCDD-treated) received TCDD (1 μ g/kg, i.p.) and saline (p.o.); groups 3 and 4 received crude saponin from Korean red ginseng (CGS) at a daily oral doses of 10 and 20 mg/kg, respectively for 4 wks; groups 5 and 6 received CGS (10 and 20 m/jkg, respectively) for 5 wks starting 1 wk before TCDD-exposure; groups 7 and 8 received CGS (10 and 20 mg/kg, respectively) for 3 wks from 1 wk after TCDD-exposure. Body weight of G2 was significant1y decreased from 2 wks after TCDD-exposure (p<0.01). Body weight of the CGS-treated groups also decreased by TCDD-exposure but the weight loss was greatly retarded compared with that of G2. From these results, we could understand the active compounds of Korean red ginseng that confront TCDD toxicity. In addition, this result further supports the preventive and therapeutic effect of Korean red ginseng against TCDD-induced toxicity.
