목적: 엘더플라워(Sambucus nigra) 추출물을 이용하여 항산화 효과, 주름개선 효능, 세포독성 평가를 실시함으로써 화장품 소재로서의 활용 가능성을 검토하였다. 방법: 항산화 활성으로는 1,1-diphenyl-2-picrylhydrazyl (DPPH)와 superoxide dismutase (SOD)-like activity, 주름개선 효능으로는 elastase 저해 활성을 측정하였다. B16-F10 mouse melanoma cells과 CCD-986sk human dermal fibroblast cells을 대상으로 세포독성 평가를 실시하였다. 결과: 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity 방법으로 실시한 항산화 활성은 시료의 농도 1000 µg/mL, 500 µg/mL, 250 µg/mL에서 Sambucus nigra water extract (SNE)가 77%, 76.4%, 75.2%의 radical 소거 활성을 나타내었으며, SOD-like activity를 측정한 결과, 시료의 농도 200 µg/mL, 100 µg/mL, 50 µg/mL에서 SNE가 78%, 70%, 66%의 활성을 나타내었다. 또한 SNE는 500 µg/mL의 농도에서 48%의 elastase 저해 활성을 나타내었다. B16-F10 mouse melanoma cells에 대한 세포 독성은 시료의 농도 200 µg/mL, 100 µg/mL, 50 µg/mL에서 각각 118%, 115%, 109%의 세포 생존율을 나타내었고, CCD-986sk human dermal fibroblast cells에 대한 세포 독성은 시료의 농도 200 µg/mL, 100 µg/mL, 50 µg/mL에서 마찬가지로 높은 세포 생존율을 확인할 수 있었다. 결론: 본 연구 결과 엘더플라워 추출물은 항산화 효과 및 주름개선 효능이 우수함을 나타내어 기능성 화장품 소재로서의 높은 활용 가능성 확인할 수 있었다.
Abstract
Purpose: This study examined the potential of elderflower (Sambucus nigra) water extract (SNE) as a cosmetic material by evaluating its anti-wrinkle, antioxidant, and cytotoxic effects. Methods: We determined the antioxidant effect of SNE by measuring its 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical–scavenging and superoxide dismutase (SOD)-like activity and determined the anti-wrinkle effect of SNE by measuring its elastase inhibition activity. In addition, we assayed the viability of B16-F10 mouse melanoma and CCD-986sk human dermal fibroblast cells in the presence of SNE. Results: SNE showed a DPPH radical–scavenging activity of 77%, 76.4%, and 75.2% at a concentration of 1000, 500, and 250 µg/mL, respectively, and SOD-like activity of 78%, 70%, and 66% at a concentration of 200, 100, and 50 µg/mL, respectively. In addition, 500 µg/mL of SNE exhibited 48% elastase inhibition activity. Furthermore, B16-F10 cells showed a viability of 118%, 115%, and 109% when treated with 200, 100, and 50 µg/mL of SNE, respectively, and CCD-986sk cells showed high cell viability when treated with 200, 100, and 50 µg/mL of SNE. Conclusion: The results of this study indicate that SNE has excellent antioxidant and anti-wrinkle effects, confirming its high applicability as a functional cosmetic material.
