Abstract
To prepare basic data for examining the potential for use of henna and indigo as natural material for cosmetics, this study investigated total polyphenol content, antioxidant, and pro-inflammatory cytokine TNF-α production inhibitory, and 5α-reductase activity inhibitory effects of a henna and indigo ethanol extract. The results showed that henna had a higher total polyphenol content (29.63 mg/g) than indigo (20.25 mg/g). The DPPH radical scavenging activity showed a concentration-dependent increase, and the antioxidant effect of henna was significantly higher (88.9%) compared with the indigo (73.8%) at the concentration of 1,000 ug/mL. Cytotoxicity for RAW264.7 cells was not observed up to the concentration of 500 μg/mL in both the henna and indigo. Production of pro-inflammatory cytokine TNF-α was significantly inhibited in both henna and indigo extracts compared with the group treated with LPS alone. Proliferation of HDP cells was observed in both henna and indigo extracts at concentrations up to 100 ug/mL and a proliferative effect was observed for henna that was twice as high as indigo. In addition, activity of 5α-reductase was inhibited by both henna and indigo. Therefore, henna and indigo ethanol extracts have potential as natural cosmetic material for anti-inflammation and prevention of hair loss. Future studies will require demonstration of the physiological activity, such as anti-inflammatory and hair loss prevention mechanisms through component analysis.
