Abstract
This study was carried out to determine the optimal condition of enzymatic hydrolysis fromacid-alkali pre-treated and steam-exploded biomass of Miscanthus sinensis var. purpurascens forethanol production. The acid-alkali pre-treated biomass contained 5% extractive, 17.0% lignin,55.1% hemicellulose and 22.7% cellulose. Saccharification using mixed enzymes was higher thanthat of single enzyme treatment. Saccharification rate by Celluclast 1.5L was highest comparedwith Viscozyme treatment. The highest saccharification was obtained in EM-3(Celluclast24.3FPU/g substrate+Viscozyme 80.6FBG/g substrate) treatment which saccharification ratereached 70%. Saccharification of purified α-cellulose was higher than that of 1% NaOH treatedsteam-exploded biomass. The optimized conditions on enzymatic hydrolysis were withtemperature of 50℃, incubation time of 48 hr. Pretreated sample contained xylose as a majorpentose, and glucose as a major hexose. Ethanol fermentation by Saccaromyces cerevisiae KCCM 11215 relies on the enzyme combination and pretreatment method. Ethanol yield in α-cellulose was highest compared to 1% NaOH and 1% KOH exploded biomass. α-cellulosesample produced the highest in ethanol yields as 350g per 1 kg biomass.
